A phylogenomic appraisal of the evolutionary relationship of mycoplasmas Genet. Mol. Biol.
Yotoko,Karla S.C.; Bonatto,Sandro L..
Several genomes of mycoplasmas have been sequenced and here we tried to retrieve the evolutionary relationships of nine species using a phylogenomic approach. Several methods were used to build phylogenetic trees based on protein sequence information, gene-order, and gene-content. We also utilized datasets composed of individual and concatenated sets of orthologous proteins, as well as with reduced unreliable alignment regions. Most of our results converge to a single topology, except for the trees built with both the maximum parsimony method and with the gene-order dataset. The gene-content dataset presented trees consistent with most nodes of the convergent tree, but in the gene-order dataset most internal branches were clearly saturated and unreliable....

ABC transporters in Mycoplasma hyopneumoniae and Mycoplasma synoviae: insights into evolution and pathogenicity Genet. Mol. Biol.
Nicolás,Marisa Fabiana; Barcellos,Fernando Gomes; Nehab Hess,Pablo; Hungria,Mariangela.
ABC transporters represent one of the largest superfamilies of active membrane transport proteins (MTPs) with a highly conserved ATPase domain that binds and hydrolyzes ATP, supplying energy for the uptake of a variety of nutrients and for the extrusion of drugs and metabolic wastes. The complete genomes of a non-pathogenic (J) and pathogenic (7448) strain of Mycoplasma hyopneumoniae, as well as of a pathogenic (53) strain of Mycoplasma synoviae have been recently sequenced. A detailed study revealed a high percentage of CDSs encoding MTPs in M. hyopneumoniae strains J (13.4%), 7448 (13.8%), and in M. synoviae 53 (11.2%), and the ABC systems represented from 85.0 to 88.6% of those CDSs. Uptake systems are mainly involved in cell nutrition and some might be...

Avian mycoplasmosis update Rev. Bras. Ciênc. Avic.
Nascimento,ER; Pereira,VLA; Nascimento,MGF; Barreto,ML.
Avian mycoplasmas occur in a variety of bird species. The most important mycoplasmas for chickens and turkeys are Mycoplasma gallisepticum (MG), M. synoviae (MS), and M. meleagridis. Besides, M. iowe (MI) is an emerging pathogen in turkeys, but of little concern for chickens. Mycoplasmas are bacteria that lack cell wall and belong to the class Mollicutes. Although they have been considered extracellular agents, scientists admit nowadays that some of them are obligatory intracellular microorganisms, whereas all other mycoplasmas are considered facultative intracellular organisms. Their pathogenic mechanism for disease include adherence to host target cells, mediation of apoptosis, innocent bystander damage to host cell due to intimate membrane contact,...

Circulation of Major Respiratory Pathogens in Backyard Poultry and their Association with Clinical Disease and Biosecurity Rev. Bras. Ciênc. Avic.
Batista,IA; Hoepers,PG; Silva,MFB; Nunes,PLF; Diniz,DCA; Freitas,AG; Cossi,MVC; Fonseca,BB.
SUMMARY Raising backyard birds is a common practice in Brazil, mainly in the countryside or suburban areas. However, the level of respiratory pathogens in these animals is unknown. We sampled two hundred chickens from 19 backyard flocks near commercial poultry farms and performed ELISA to Infectious Bronchitis Virus, avian Metapneumovirus, Mycoplasma synoviae and Mycoplasma gallisepticum. We evaluated the association between the predictive ability of ELISA and Hemagglutination-inhibition (HI)by comparing results from eight flocks positive to Mycoplasma gallisepticum on ELISA. Besides, we assessed essential biosecurity measures in the properties (multiple species birds, rodent control, hygienic conditions, and water quality for the bird`s consumption). We...

Clastogenic effects of different Ureaplasma urealyticum serovars on human chromosomes BJMBR
The possibility that Ureaplasma urealyticum might play an important role in human infertility was first raised more than 20 years ago, but this association remains speculative. Considering the hypothesis that the pathogenicity of Ureaplasma urealyticum may depend on its serotypes, the clastogenic effects of different strains of Ureaplasma urealyticum, at concentrations of 103 CCU (color changing units)/ml, 104 CCU/ml and 105 CCU/ml, were evaluated in vitro in short-term cultures of human lymphocytes. Total or partial mitotic inhibition was produced by Ureaplasma urealyticum serotypes 2, 3 and 10 independent of the concentration (103 CCU/ml, 104 CCU/ml or 105 CCU/ml) of the microorganisms employed. In contrast, the clastogenic effects observed with...

Comparative genome analysis of proteases, oligopeptide uptake and secretion systems in Mycoplasma spp Genet. Mol. Biol.
Staats,Charley Christian; Boldo,Juliano; Broetto,Leonardo; Vainstein,Marilene; Schrank,Augusto.
Mycoplasmas are very fastidious in their nutritional requirements for in vitro growth and have limited biosynthetic capacity, a reflection of their reduced genomes. As a result, these bacteria depend upon external metabolites for nutrition and growth and have developed dependence on their hosts for survival and maintenance. Protein degradation and peptide importation play an important role in Mycoplasma spp. nutrition, and proteases can play a role in host adaptation and pathogenicity. Here, we present a general survey on the genes involved in protein degradation, secretion and importation, comparing all available Mollicute genomes.

Intracellular microcolonies of bacteria (IMC), in some cases developing large extracellular cysts (bacterial aggregates), infecting primarily gill and digestive gland, have been historically reported in a wide diversity of economically important mollusk species worldwide, sometimes associated with severe lesions and mass mortality events. As an effort to characterize those organisms, traditionally named as Rickettsia or Chlamydia-like organisms, 1950 specimens comprising 22 mollusk species were collected over 10 countries and after histology examination, a selection of 99 samples involving 20 species were subjected to 16S rRNA gene amplicon sequencing. Phylogenetic analysis showed Endozoicomonadaceae sequences in all the mollusk species analyzed....

Description of Lyme disease-like syndrome in Brazil: is it a new tick borne disease or Lyme disease variation? BJMBR
Mantovani,E.; Costa,I.P.; Gauditano,G; Bonoldi,V.L.N.; Higuchi,M.L.; Yoshinari,N.H..
An emerging clinical entity that reproduces clinical manifestations similar to those observed in Lyme disease (LD) has been recently under discussion in Brazil. Due to etiological and laboratory particularities it is named LD-like syndrome or LD imitator syndrome. The condition is considered to be a zoonosis transmitted by ticks of the genus Amblyomma, possibly caused by interaction of multiple fastidious microorganisms originating a protean clinical picture, including neurological, osteoarticular and erythema migrans-like lesions. When peripheral blood of patients with LD-like syndrome is viewed under a dark-field microscope, mobile uncultivable spirochete-like bacteria are observed. PCR carried out with specific or conservative primers to recognize...

Detection of multiple mycoplasma infection in cell cultures by PCR BJMBR
Timenetsky,J.; Santos,L.M.; Buzinhani,M.; Mettifogo,E..
A total of 301 cell cultures from 15 laboratories were monitored for mycoplasma (Mollicutes) using PCR and culture methodology. The infection was detected in the cell culture collection of 12 laboratories. PCR for Mollicutes detected these bacteria in 93 (30.9%) samples. Although the infection was confirmed by culture for 69 (22.9%) samples, PCR with generic primers did not detect the infection in five (5.4%). Mycoplasma species were identified with specific primers in 91 (30.2%) of the 98 samples (32.6%) considered to be infected. Mycoplasma hyorhinis was detected in 63.3% of the infected samples, M. arginini in 59.2%, Acholeplasma laidlawii in 20.4%, M. fermentans in 14.3%, M. orale in 11.2%, and M. salivarium in 8.2%. Differential metabolism of Mycoplasma species as revealed by their genomes Genet. Mol. Biol.
Arraes,Fabricio B.M.; Carvalho,Maria José A. de; Maranhão,Andrea Q.; Brígido,Marcelo M.; Pedrosa,Fábio O.; Felipe,Maria Sueli S..
The annotation and comparative analyses of the genomes of Mycoplasma synoviae and Mycoplasma hyopneumonie, as well as of other Mollicutes (a group of bacteria devoid of a rigid cell wall), has set the grounds for a global understanding of their metabolism and infection mechanisms. According to the annotation data, M. synoviae and M. hyopneumoniae are able to perform glycolytic metabolism, but do not possess the enzymatic machinery for citrate and glyoxylate cycles, gluconeogenesis and the pentose phosphate pathway. Both can synthesize ATP by lactic fermentation, but only M. synoviae can convert acetaldehyde to acetate. Also, our genome analysis revealed that M. synoviae and M. hyopneumoniae are not expected to synthesize polysaccharides, but they can take...

Disturbance induced decoupling between host genetics and composition of the associated microbiome ArchiMer
Wegner, Karl Mathias; Volkenborn, Nils; Peter, Hannes; Eiler, Alexander.
Background Studies of oyster microbiomes have revealed that a limited number of microbes, including pathogens, can dominate microbial communities in host tissues such as gills and gut. Much of the bacterial diversity however remains underexplored and unexplained, although environmental conditions and host genetics have been implicated. We used 454 next generation 16S rRNA amplicon sequencing of individually tagged PCR reactions to explore the diversity of bacterial communities in gill tissue of the invasive Pacific oyster Crassostrea gigas stemming from genetically differentiated beds under ambient outdoor conditions and after a multifaceted disturbance treatment imposing stress on the host. Results While the gill associated microbial communities in...

Economic Consequences of Biological Variation: The Case of Mycoplasma in Swine Herds AgEcon
Otto, Lars.
We present an economic decision support model, based on a Bayesian network, for Mycoplasma infection in slaughter swine production. The model describes the various risk factors for Mycoplasma infection and their interactions. This leads to a stochastic determination of the consequences of productivity factors, and this again results in stochastic economic consequences when changing the risk factors or engaging in control arrangements. We use the model to calculate how estimated prevalence and level of Mycoplasma changes when we gather evidence from various veterinarian examinations, and we show how this influences the distribution of the economic results of a change in strategy to fight Mycoplasma. One result is that the profitability in fighting...

Hemoparasitosis, diseases that may cause clinical and hematological complications, are considered a great challenge to practitioners and clinical pathologists. Its high incidence in all Brazilian territory and the difficulty for a precise diagnosis can lead in most cases to a therapeutic diagnosis. Anaplasma platys, Babesia canis vogeli, Ehrlichia canis, Mycoplasma haemocanis and with a lower frequency and clinical relevance, Anaplasma phagocytophilum are the main etiological agents affecting dogs in Brazil. The climatic conditions found in several Brazilian areas, such as high rain and temperature levels, favor the arthropods vectors life cycles. This subject became an important challenge to both veterinarians and pet owners. These etiological agents...

Identification of the GTPase superfamily in Mycoplasma synoviae and Mycoplasma hyopneumoniae Genet. Mol. Biol.
Borges,Clayton Luiz; Parente,Juliana Alves; Pereira,Maristela; Soares,Célia Maria de Almeida.
Mycoplasmas are the smallest known prokaryotes with self-replication ability. They are obligate parasites, taking up many molecules of their hosts and acting as pathogens in men, animals, birds and plants. Mycoplasma hyopneumoniae is the infective agent of swine mycoplasmosis and Mycoplasma synoviae is responsible for subclinical upper respiratory infections that may result in airsacculitis and synovitis in chickens and turkeys. These highly infectious organisms present a worldwide distribution and are responsible for major economic problems. Proteins of the GTPase superfamily occur in all domains of life, regulating functions such as protein synthesis, cell cycle and differentiation. Despite their functional diversity, all GTPases are believed to have...

Insertion sequences as variability generators in the Mycoplasma hyopneumoniae and M. synoviae genomes Genet. Mol. Biol.
Loreto,Elgion Lúcio Silva; Ortiz,Mauro Freitas; Porto,Jorge Ivan Rebelo.
We have analyzed the sequenced genomes of three strains of Mycoplasma hyopneumoniae and one strain of M. synoviae, and have found three and two different transposable element families, respectively in each species. In M. hyopneumoniae, the Insertion Sequences of the IS4 family is represented by ISMHp1, a putatively active element. The IS3 family is represented by several degenerated sequences. A third element called tMH was found, which shows some characteristics reminiscent of retrotransposons. In M. synoviae, three different possibly active IS4 elements are present (ISMHp1-like; ISMs1 and IS1634-like elements). The IS30 family is represented by the degenerated IS1630-like element. The IS1634-like element is shown to be involved in chromosomal...

Intra-uterine experimental infection by Ureaplasma diversum induces TNF-α mediated womb inflammation in mice Anais da ABC (AABC)
Silva,Jamile R.; Ferreira,Lício F.A.A.; Oliveira,Percíllia V.S.; Nunes,Ivanéia V.; Pereira,Ítalo S.; Timenetsky,Jorge; Marques,Lucas M.; Figueiredo,Tiana B.; Silva,Robson A.A..
Ureaplasma diversum is an opportunistic pathogen associated with uterine inflammation, impaired embryo implantation, infertility, abortions, premature birth of calves and neonatal pneumonia in cattle. It has been suggested that the intra-uterine infection by Ureaplasma diversum can cause vascular changes that hinder the success of pregnancy. Thus, the aim of this study was to evaluate the changes of intrauterine site of A/J mice in estrus or proestrus phase inoculated with Ureaplasma diversum. The infection was monitored at 24, 48 and 72 hours by the PCR methodology to detect the Ureaplasma in the inoculation site and the profile of circulating blood cells. Morphological changes, intensity of inflammation and the production of cytokines were compared. The...

Kinases of two strains of Mycoplasma hyopneumoniae and a strain of Mycoplasma synoviae: an overview Genet. Mol. Biol.
Bailão,Alexandre Melo; Parente,Juliana Alves; Pereira,Maristela; Soares,Célia Maria de Almeida.
Mycoplasma synoviae and Mycoplasma hyopneumoniae are wall-less eubacteria belonging to the class of Mollicutes. These prokaryotes have a reduced genome size and reduced biosynthetic machinery. They cause great losses in animal production. M. synoviae is responsible for an upper respiratory tract disease of chickens and turkeys. M. hyopneumoniae is the causative agent of enzootic pneumonia in pigs. The complete genomes of these organisms showed 17 ORFs encoding kinases in M. synoviae and 15 in each of the M. hyopneumoniae strain. Four kinase genes were restricted to the avian pathogen while three were specific to the pig pathogen when compared to each other. All deduced kinases found in the non pathogenic strain (J[ATCC25934]) were also found in the...
